Summary: | This thesis work will approach the topic of pancreatic ductal adenocarcinoma (PDA) intractability in two distinct parts. The first chapter (Chapter 1; Pages 16–86) will cover the interplay between the systemic immune system, aerobic exercise, and tumor growth in pancreatic cancer. Focusing specifically on the potential anti-tumor and immune-activating effects of aerobic exercise, the work herein will describe the therapeutic potential and mechanisms that underlie an exercise-oncology axis in this disease. The second chapter (Chapter 2; Pages 87–145) will approach the topic of aberrant glycosylation in PDA, an emerging hallmark of cancer that contributes to both treatment resistance and immune-suppression. In particular, the work herein will use lectin array technology detecting glycosylation-epitopes in both human and murine tissues to identify glyco-related genes that promote tumor growth. In aggregate, this work highlights the contribution of systemic immune mobilization to PDA anti-tumor immunity and underscores the contribution of glycosylation to PDA development. |
