Summary: | Dioecy, the separation of male and female organs into different individuals, is rare in flowering plants. Populus nigra, the black cottonwood tree, is a member of the genus Populus, in which all member species are dioecious. In Populus species, the mechanism for sex determination is not known, though there is evidence that Populus nigra specifically carries an XY determination system. We used existing sequence data to investigate the sex-specific region of the Populus nigra Y chromosome. Using a restriction enzyme based method, we determined the sex of a population of 700+ half sibling seedlings that were too young to sex phenotypically. Male and female genomic sequence read pools were searched for male sex-specific sequences by extracting read subsequences of length k, or k-mers, a method that has been used previously in the discovery of the sex-determining mechanism of persimmon. The sex-specific sequences were then used to assemble male-specific regions. Samples of the assembled sequences were verified by PCR, thus validating this method as a preliminary approach for identifying sex-specific sequences. |
