Summary: | In mushrooms in Italy viruslike particles of the following four types were found : a) isometric particles with diameters mostly 40-42 nm ; b) isometric particles with diameter 28-30 nm ; c) bacilliform particles of about 55 × 20 nm and d) rod-shaped particles 18-19 nm wide. All these particles were partially purified by extracting sap from diseased mushrooms in the presence of buffers and protective solutions, then by clarifying the extracts with chloroform or n-butanol and finally by concentrating the particles with two differential centrifugation cycles before and after a sucrose gradient ultracentrifugation ; this last improved the purification and permitted the separation of some types of particles, which were together in the original preparation. In funghi coltivati in Italia sono state rinvenute particelle virus-simili di (iiiattro tipi : tondeggianti con diametro prevalente di 40-42 nm, tondeggianti con diametro di 28-30 nm, bacilliformi di circa 55 × 20 nm e a bastoncino di 18-19 nm di larghezza. Tali particelle sono state purificate parzialmente estraendo il succo dai funghi infetti in presenza di tamponi e sostanze protettrici, chiarificando tali estratti con cloroformio o n-butanolo e precipitando le particelle con 2 cicli di centrifugazione differenziale, inframmezzati da una ultracentrifugazione in gradiente di densità di saccarosio; quest'ultima ha migliorato la purificazione ed ha anche consentito di separare alcuni tipi di particelle presenti insieme nella preparazione di origine. |
