Single nucleotide polymorphisms of adiponectin gene in breast cancer

INTRODUCTION: Polymorphisms in adiponectin gene [(rs1501299 (G276T) and rs266729 (C11377G)] are known to influence plasma levels of adiponectin. Literature is scarce on the association of adiponectin gene polymorphism rs266729 with breast cancer. The present study was taken up to study these polymorphisms and their association with breast cancer. MATERIALS AND METHODS: Ninety three patients diagnosed with malignant breast cancer were included as cases along with 186 age matched healthy controls. Adiponectin rs1501299 (276G/T) and rs266729(-11377 C/G) were studied using polymerase chain reaction (PCR) based restriction fragment length polymorphism (RFLP). RESULTS: Adiponectin rs1501299 (276G/T) showed a significant 2.6-fold increased risk for breast cancer [OR 2.6,95% CI 1.575-4.396)] No association was found with Adiponectin rs 266729(-11377 C/G). Further, presence of SNP in adiponectin gene rs 1501299 was associated with breast cancer in both obese and non obese patients as well as in pre and post menopausal patients with breast cancer. No association was seen with rs 266729 with breast cancer. CONCLUSION: Adiponectin 276G/T is associated with breast cancer irrespective of irrespective of BMI and the menopausal state in south Indian subjects. No such association was seen with adiponectin gene -11377C/G. KEY WORDS: Adiponectin, SNP, rs1501299, rs266729,
