Summary: | [Myostatin is a protein codified by GDF8 gene which operates as a negative regulator of muscle growth. In several cattle breeds the occurrence of a not properly working myostatin, often as a consequence of mutations in the third exon, results in "double muscled" phenotype. In some breeds the selection caused the fixation of mutated alleles (Belgian Blue, Piemontese), while in Marchigiana the frequency of such alleles is low. In the present work we have estimated the onset time of the mutation in such breeds by utilizing microsatellite molecular markers in linkage within the same myostatin chromosome (2). The estimated times of onset of the mutations resulted very short in all breeds, particularly the one carried by Marchigiana. In this breed the linkage disequilibrium is still present in a large part of chromosome 2.]
La miostatina è una proteina codificata dal gene GDF8 che funziona come regolatore negativo della crescita muscolare. In diverse razze bovine è stato visto che la presenza di una miostatina malfunzionante, spesso in conseguenza a mutazioni nel terzo esone, causa il fenotipo "doppia coscia". In alcune razze la selezione ha provocato la fissazione degli alleli mutati (Blu Belga, Piemontese), mentre nella Marchigiana la frequenza di tali alleli è limitata. Nel presente lavoro abbiamo datato la comparsa della mutazione in tali razze utilizzando marcatori molecolari microsatelliti in linkage sullo stesso cromosoma della miostatina (2). Tutte e tre le razze risultano aver acquisito la rispettiva mutazione piuttosto recentemente, in particolare la Marchigiana.
