Summary:Selenocysteine tRNA serves as the carrier molecule for the biosynthesis of selenocysteine. Two distinct isoacceptors of this tRNA, designated tRNA super((Ser)Sec), have been observed in higher eukaryotes. Despite the existence of these two related tRNAs, only a single hybridizing locus is detected in bovine DNA. In order to isolate this locus, a recombinant bovine/EMBL 3 library was screened by hybridization with a human selenocysteine tRNA super((Ser)Sec) gene probe. A total of six positively hybridizing phage were isolated and determined to represent two, overlapping segments of genomic DNA. The hybridizing segments within both clones contained a 3.0 Kb EcoRI fragment also detected in genomic DNA, thus indicating the isolation of the entire hybridizing region of bovine DNA.
