Abstract A simple spectroscopic method is proposed to control the "ripening" of Delafield's hematoxylin solution during natural "ripening" or fast oxidation by different agents. The hematoxylin solution has ripened sufficiently for use with tissue sections staining if the absorption is 1.2-1.3 at 560 nm. The hematoxylin solution must first be diluted 15-fold using a 5% ammonium alum solution and the absorption must be measured in 0.5 cm cuvettes.
