Abstract Patients with cholangiocarcinoma (CCA) tumor located in the biliary tract, have low survival due to difficulties in diagnosis in its early stage. In this context, microRNAs with inhibitory potential on tumor proliferation, such as miR-101-3p, are a potential molecular marker for diagnosis and prognosis of CCA. The aim of this study was to evaluate the expression of miR-101-3p in patients with CCA and its relationship with survival, lifestyle habits and comorbidities. 41 patients (SG) (median age: 56 y; 51.2% female) and 21 individuals from the control group (CG) (median age: 39 y; 80.9% female) who underwent cholecystectomy were studied. RNA was extracted from tumor tissue embedded in paraffin in SG and fresh tissue in CG. Expression analysis of miR-101-3p was performed by real-time reverse transcription polymerase chain reaction with significance level assumed for a value of p<0.05. The analysis of miR-101-3p expression showed that it was underexpressed in the SG compared to the CG (0.17; p=0.0001), which didn't occur for lifestyle habits and comorbidities (p>0.05). For lifestyle habits, smoking prevailed in patients (36.6%; p=0.034) while alcoholism was similar in both groups (24.4%, 23.8%; p>0.05). The same occurred for systemic arterial hypertension (24.4%; 23.8%, p>0.05, respectively) and diabetes mellitus (9.75%, 19.0%, p>0.05, respectively). However, body mass index (BMI) ≥25kg/m² excelled in controls (66.6%, p=0.012). The underexpression of mir-101- 3p, an important tumor suppressor, is associated with patients with CCA. Smoking and increasing age seem to be important independent risk factors for cholangiocarcinoma.
