Summary:S-haplotypes and self-compatibility of the Japanese plum cultivar 'Karari' were investigated. Using the PCR method with S-RNase gene-specific primers, the S-haplotype of 'Karari' was determined to be SbSt. Partial genomic DNA for a new S-RNase gene (St) was cloned and sequenced. Self-pollinated flowers of 'Karari' set fruit, confirming that this cultivar is self-compatible. The progenies of the self-pollination segregated into three S-haplotypes, SbSb, SbSt and StSt. Thus, both the Sb- and the St-haplotypes may be responsible for self-compatibility.
