Summary: | The role of differential post-transcriptional processing of globin mRNA in the accumulation of this RNA during murine erythroleukemia cell differentiation has been evaluated by two approaches. First, the incorporation of [
3H]uridine into globin RNA was followed throughout induction after short and long exposures to the isotope. Incorporation after short times is an index of globin gene transcription, while that after longer times reflects both synthesis and post-transcriptional events. A comparison of the incorporation at these two times will reveal differential processing. The second approach was to determine the proportion of pulse-labeled RNA found in mature globin mRNA and each of its precursors throughout induction. These proportions are a measure of precursor processing rate and this approach demonstrates whether regulation occurs on the level of precursor processing or stabilization. No differential processing was seen by either of these approaches and we conclude that transcriptional activation of the globin gene is the primary mechnism for globin mRNA accumulation in inducing murine erythroleukemia cells. |
