Summary:Anti-ulcer and anti-secretory effects of an inclusion-complex of 4-t-butyl-2'-carboxy-methoxy-4'-(3-methyl-2-butenyloxy) chalcone (SU-740), which inhibits prostaglandin (PG) degradation and increases PGs contents in gastric mucosa, were studied in rats. SU-840 is the inclusion-complex of SU-740 with β-cyclodextrin. SU-740 and SU-840 given intraduodenally showed a remarkable inhibition of 0.6N HCl-induced gastric lesions.The protective effect of SU-740 was potentiated about 5 times by inclusion with β-cyclodextrin. SU-840 showed a marked anti-ulcer effect in various experimental ulcer models. The anti-ulcer effect of SU-840 was significantly attenuated by pretreatment with indomethacin in the 0.6N-HCl induced gastric lesion model. Gastric secretion was also inhibited by SU-840 in pylorus-ligated rats but the anti-secretory effect of SU-840 was not inhibited by pretreatment with indomethacin. These results suggest that SU-840 shows an anti-ulcer effect with at least two different mechanisms:one is mediated by PG and the other is not mediated by PG.
