Anim. Sci. 91: 255-264. Pregnancy rates (PR) in high-producing lactating dairy cows have declined drastically over the past several decades, but those of heifers have remained constant. Reduced PR could be due to multiple causes, and the underlying pathophysiological mechanisms are still unclear. A compromised maternal uterine environment could be one of factors that could affect the PR. This study was performed to compare the nature of the uterine environment in dairy heifers and lactating dairy cows (2nd/3rd parity) by analyzing the expression levels of selected endometrial genes. Estrus was synchronized in heifers (n=5) and lactating dairy cows (n=5) and endometrial biopsies were performed during the mid luteal phase (day 11) of the estrous cycle. Real-time polymerase chain reaction (Q-RT PCR) and immunohistochemistry were performed to analyse the mRNA and protein levels of genes respectively. Relative abundance of BCL2, HSPA1A, IL1A, TNF, IGF1, FGF2 and SERPINA14 transcripts and the protein expression of IL1A, TNF and FGF2 were significantly higher in heifers in comparison with lactating dairy cows. Our findings suggest an altered endometrial environment in lactating dairy cows compared with heifers. However, whether these differences play a role in pregnancy outcomes should be further investigated.

Comparaison du taux d'expression de certains gènes de l'endomètre chez les génisses laitières et les vaches laitières en lactation. Can. J. Anim. Sci. 91: 255-264. Le taux de gravidité (TG) des vaches laitières produisant beaucoup de lait a considérablement diminué au cours des dernières décennies, tandis que celui des génisses laitières est demeuré le même. La baisse du TG pourrait résulter de causes multiples, et on connaît mal les mécanismes physiopathologiques sous-jacents. Une modification de l'environnement utérin chez la mère pourrait faire partie des facteurs expliquant le plus faible TG. La présente étude devait comparer l'environnement utérin des génisses laitières et des vaches laitières en lactation (2e/3e rang de mise bas) par analyse du taux d'expression des gènes de l'endomètre. Les auteurs ont synchronisé l'æstrus des génisses (n=5) et des vaches (n=5), puis procédé à une biopsie de l'endomètre au milieu de la phase lutéale (11ejour) du cycle. Ils ont analysé l'ARNm et l'expression des protéines des gènes, respectivement, par la réaction en chaîne de la polymérase en temps réel (Q-RT PCR) et par immunohistochimie. L'abondance relative des transcrits de BCL2, HSPA1A, IL1A, TNF, IGF1, FGF2 et SERPINA14 et l'expression des protéines IL1A, TNF et FGF2 étaient sensiblement plus élevées chez les génisses. Ces résultats laissent croire que l'endomètre des vaches laitières en lactation est différent de celui des génisses. D'autres études devraient être entreprises pour établir si les variations notées jouent un rôle dans l'issue de la gestation.
