Summary: | This work aimed to optimize the in vitro multiplication of (L.) Willk. Gorse is a shrub naturally distributed in the north and center of the Iberian Peninsula and in North Africa. It is intensively used in popular medicine and gastronomy and also as an ornamental. The plant material established in vitro was collected in three different geographical areas in Portugal: Malcata, Gardunha and Orvalho. In the multiplication stage we compared eight culture media with different growth regulators and nitrates composition. The parameters measured were number of shoots, shoot length and number of segments per treatment (culture media). The Murashige and Skoog medium, with nitrates full strength supplemented with 1 mg/L 6-benzylaminopurine (BAP) and 0.5 mg/L indole-3-butyric acid (IBA) revealed to be the best medium for shoot growth and proliferation. The Orvalho material displayed the highest number of shoots (10.4) and segments (43.9) and the Malcata had the best elongation (6.8 cm). Reduction of nitrates in the medium had no influence on the different studied parameters. No hyperhydricity was observed in shoots grown on both type of media, half and full-strength nitrate composition. The results revealed that all the media used displayed high multiplication rates, which guarantee a cost effective multiplication system for the species. |
