Summary:We analyzed the potential of a polyphenolic plant metabolite Gallic Acid (GA) for treating gastric cancer. AGS gastric cell line was exposed to varying concentrations of GA at several intervals, and treatment efficiency was assessed via DNA microarray gene expression signature, quantitative RT-PCR, and immunocytochemistry followed by confocal microscopy. We specifically focused on detecting the changes in Epithelial to Mesenchymal Transition (EMT) markers, indicative of metastatic and treatment resistant cancers. A 48 hour exposure to 100 micromolar solution of GA resulted in over 50% decrease in cells with EMT morphology, decrease of mesenchymal marker Vimentin, and increase of epithelial marker E-Cadherin. Our data suggests that GA exposure can help reverse the transformation of cancerous cells, thus aiding in faster and more efficient treatment.
